Hot Water Extract of Perilla frutescens var. acuta Regulates Contraction in the Primary Cultured Bovine Ciliary Smooth Muscle Cells via the PLC/IP3 and cAMP/PKA Pathways

Abstract
We investigated the regulatory effects of a hot water extract of Perilla frutescens var. acuta (PF) on the contraction of primary cultured bovine ciliary muscle cells via the PLC/IP3 and cAMP/PKA pathways. In this study, we treated cyclopentolate hydrochloride (10 ¥ìM) on the cells to inhibit ciliary muscle contraction and then we treated these cells with PF (50 or 200 ¥ìg/mL) to restore the accommodation process of these the cells. Intracellular levels of IP3, Ca2+ and cAMP were measured by ELISA. For determining the mode of action in the signaling pathways, the protein expressions of IP3, CaM, MLCK and PKA were measured by performing western blotting. As a result, PF increased the intracellular level of Ca2+ and CaM, promoted by IP3 activation, indicating that certain components of PF bind to the G-protein coupled receptor (GPCR) and so activate the PLC/IP3 signaling pathway. On the other hand, the PF elevated the intracellular levels of cAMP and PKA, and this resulted in the activation of MLCK to regulate ciliary muscle contraction. These results suggested that PF has an ability to regulate ciliary muscle contraction via both the PLC/IP3 and cAMP/PKA signaling pathways in cyclopentolate hydrochloride-treated primary cultured ciliary muscle cells. However, due to characteristics of ciliary smooth muscle, this study carefully suggests that PF is more favorable to the cAMP/PKA pathway rather than the PLC/IP3 pathway in contraction or restoring the accommodation process via further activation of MLCK.
